I am curently integrating reach with Navio2. I have connected the rover reach with navio2. Do i really need a 3DR radio UART connection between the two reach device to establish the connection. And if not then how do I integrate Navio2 with reach ?

No, you can use Mavlink inject for RTCM corrections. Just connect Reach to Navio, configure it as second GPS and most of the things from this guide are applicable to Navio https://docs.emlid.com/reach/apm-integration/ .
