Is it possible to use Reach to find position and heading between two drones?
if it is possible, what is the best way to do it?

One stationary base and two Reach receivers on the drones. You will need some simple distance and heading calculations after that.

Dose Reach support multiple rovers connected to one base or should I use one base for each rover?

Yes, you can use virtually unlimited number of rovers with one base.
